Unusual amino acids, particularly β-, γ-, and other non-α variants, have attracted considerable attention in recent yearsduetotheir applications in drug discovery, peptide-based therapeutics, and materials science. These modified amino acids exhibit uniqueconformational flexibility, enhanced stability against enzymatic degradation, and the ability to mimic protein secondarystructures.Thispaper discusses the synthetic strategies employed for generating such amino acids, including asymmetric synthesis, multicomponentreactions, and chemoenzymatic methods. Special focus is given to the synthesis of β-amino acids, their stereochemical challenges,andtheir integration into bioactive compounds.
